you can have as many USB devices for the same router as you like. As long as you have the necessary img files on them, the OpenWRT router will recognize them and the PB will boot up and work.
According to my understanding, FAT32 file system is used for storing uploaded files because of its compatibility across operating systems. I am not familiar with OS X, but a quick read at Wikipedia says:
Looking at file system performances at http://wiki.openwrt.org/doc/hardware/performance there is only really a difference between FAT32 and Ext4 in terms of writing speed.
More important can be the use of a good USB drive http://wiki.openwrt.org/inbox/benchmark.usb that could boost your system's performance.
I would personally suggest trying out both. If you are happy with the performance of FAT32 ( think it should be fine for streaming media etc. for just 1-2 devices), then it's fine. Why change? Try with other file system and see if it works. If yes, you have an option and can decide what to do.
It's not an official method, but just by simply copying over all files from a properly installed PB USB to another USB, it should work.
